The "Happy Hormones"

1. Dopamine - the "reward/motivation" chemical

4. Endorphins - the body's natural painkillers

Endorphins (notably beta-endorphin) are endogenous opioids that bind mu-opioid receptors to produce analgesia and euphoria (Rosen's Emergency Medicine, Table 6.2). They're released during exercise ("runner's high"), stress, laughter, and pain, and they work partly by triggering downstream dopamine release, which is why endorphins and dopamine are often described as working together (Cleveland Clinic).

How they interact

These four don't work in isolation - massage, for example, has been shown to raise dopamine, serotonin, and oxytocin simultaneously, and endorphin release upstream triggers dopamine release. Deficiencies in these systems are implicated in depression, chronic pain syndromes, and social/attachment disorders.

A note on recent evidence

A 2025 systematic review and meta-analysis in JAMA Psychiatry (PMID: 40498519) refined the classic dopamine/serotonin story, finding dopamine tracks reward processing while serotonin is more linked to punishment/aversive processing - a more nuanced picture than the simple "happy hormone" framing suggests.
