What are Primordial Germ Cells (PGCs)?

Think of PGCs as the "starter cells" for all eggs and sperm in your body.
  • They are pluripotent - meaning they have the potential to become many different types of cells (not just sex cells).
  • They are the ancestors of both male gametes (sperm) and female gametes (eggs).

Where do they come from and where do they go?

Here's the journey in simple steps:
  1. Week 2 of development: PGCs are made from a layer of cells called the epiblast (the early embryonic disc).
  2. Week 4: These cells travel to (migrate into) the yolk sac - a small sac attached to the early embryo that provides nutrition.
  3. Week 5: From the yolk sac, they migrate further and reach the genital ridge - which is basically the future site of the gonads (ovaries or testes).
The diagram in the image shows this journey: PGCs start near the allantois (another early structure), and move toward the genital ridge.

What happens when things go wrong? (Clinical Correlation)

If PGCs accidentally migrate to the wrong place during development, they can form abnormal growths called teratomas. These are tumors that can contain weird mixtures of tissues (like hair, teeth, bone) because PGCs have the potential to form many cell types.
Two classic examples:

1. Craniopharyngeal Teratoma

  • Happens when PGCs abnormally migrate up to the neck/head region.
  • Forms a tumor near the brain (craniopharyngeal area).
  • The image on the upper left shows what this looks like on a scan.

2. Sacrococcygeal Teratoma

  • Happens when PGCs wander down to the sacrum and coccyx (the lower back / tailbone area), OR when remnants of the primitive streak (another early embryonic structure) persist there.
  • This is actually the most common teratoma in newborns.
  • The image on the lower right shows a sacrococcygeal teratoma - you can see a large mass at the base of the baby's spine.
