What is the arrangement of the middle lamella, primary cell wall, secondary cell wall and the cell membrane?

From outermost to innermost, the arrangement is:
  1. Middle lamella - the outermost layer, shared between adjacent cells, composed mainly of pectin. It acts as a cement that holds neighbouring cells together.
  2. Primary cell wall - deposited inside the middle lamella during cell growth; relatively thin and flexible, made of cellulose, hemicellulose, and pectin.
  3. Secondary cell wall - present in some cell types (e.g. xylem, fibres); deposited inside the primary wall after the cell stops growing; thick, rigid, often lignified. It typically has three sub-layers (S1, S2, S3).
  4. Cell membrane (plasma membrane) - the innermost boundary, lying just inside the cell wall, separating the wall from the cytoplasm.
So the order from outside to inside is:
Middle lamella → Primary cell wall → Secondary cell wall → Cell membrane → Cytoplasm
A few points worth noting:
  • Not all cells have a secondary cell wall; it is present mainly in cells that need mechanical strength or conduct water.
  • The middle lamella is technically a shared zone between two cells, so it sits at the interface rather than belonging exclusively to one cell.
  • In woody tissues, the secondary wall is often the thickest layer and dominates the overall wall structure.
