Papez circuit is a limbic pathway involved mainly in episodic memory formation and emotional processing.

Key functions

Clinical relevance

Damage anywhere in the circuit can cause anterograde amnesia:
  • Mammillary body injury, classically in thiamine deficiency and Wernicke-Korsakoff syndrome
  • Fornix lesions
  • Anterior thalamic infarction
  • Hippocampal damage, such as from hypoxic injury or medial temporal lobe pathology
The circuit begins with hippocampal/subicular output and travels through the fornix to the mammillary bodies, then through the mammillothalamic tract to the anterior thalamic nuclei. The thalamus projects to cingulate cortex, which returns toward the parahippocampal region via the cingulum, completing the loop. Relationship

The Papez circuit is part of the limbic system, but the two are not identical.
  • Think of the limbic system as the broad network.
  • Think of the Papez circuit as one major anatomical pathway within that network, especially relevant to memory.
A key distinction is that the amygdala, central to fear, threat processing, and emotional salience, is generally included in the limbic system but is not part of the classic Papez circuit.
Modern accounts also caution that the limbic system is not a single, sharply bounded “emotion center.” Emotional behavior and memory depend on distributed networks. The Papez pathway remains a useful neuroanatomical model of medial temporal lobe-diencephalic memory connections, as summarized in this review of the updated circuit.
